Anatomy of somatic embryogenesis in Carica papaya L. BABT

Mature zygotic embryos of Carica papaya L. ‘Sunrise Solo’ were used as explants for embryogenesis induction. The explants were inoculated on Murashige and Skoog culture medium supplemented with 2 mg.L-1 2,4-dichlorophenoxyacetic acid and incubated in darkness at 25+2°C. Histological analysis of callogenesis and somatic embryogenesis indicated occurrence of direct and indirect somatic embryogenesis development. Assessment of somaclonal variation in indirect morphogenesis-derived plants of Arracacia xanthorrhiza PAB

Abstract: The objective of this work was to induce and detect somaclonal variation in arracacha (Arracacia xanthorrhiza) plants regenerated via indirect morphogenesis, in order to evaluate the potential of this technique to produce new genotypes for breeding purposes of this crop. Calli were induced from petiole segments on Murashige & Skoog (MS) medium supplied with 0.1 mg L-1 2,4-dichlorophenoxyacetic acid. The regeneration of plants via indirect morphogenesis was carried out on half-strength MS medium without plant growth regulators. Biochemical and morphological changes during the growth kinetics of Araucaria angustifolia suspension cultures BABT

Embryogenic cultures of Araucaria angustifolia were established in a BM liquid medium supplemented with 2 µM 2,4-dichlorophenoxyacetic acid, 1 µM 6-benzylaminopurine and 1 µM kinetin (BM2) and in a BM medium free of growth regulators (BM0). During 42 days in culture, the cell growth pattern of both cultures was similar. The pH of the culture medium of both BM0 and BM2 underwent progressive reduction during culture time. For both the embryogenic cultures a preferential uptake of glucose in the late stages of cell growth kinetics was observed. The objective of this study was to establish cell suspension culture and plant regeneration via somatic embryogenesis of a Brazilian plantain, cultivar Terra Maranhão, AAB. Immature male flowers were used as explant source for generating highly embryogenic cultures 45 days after inoculation, which were used for establishment of cell suspension culture and multiplication of secondary somatic embryos. Five semisolid culture media were tested for differentiation, maturation, somatic embryos germination and for plant regeneration. Two somatic embryogenesis receptor-like kinase genes (identified as AcSERK1 and AcSERK2) have previously been characterized from pineapple (Ananas comosus). In this work, we describe the characterization of a third gene (AcSERK3) in this family. AcSERK3 had all the characteristic domains and shared extensive sequence homology with other plant SERKs. AcSERK3 expression was studied by in situ hybridization and quantitative real-time PCR to analyze its function. Intense in situ hybridization signals were observed only in single competent cells and competent cell clusters; no hybridization signal was detected in the subsequent stages of somatic embryogenesis. ABSTRACT: Plants of Coffea arabica Icatu IAC 925 germplasm show resistance to Meloidogyne paranaensis under field conditions, but with variable rates of segregation. One mother plant, IAC 3, of Icatu IAC 925 was evaluated under controlled conditions through the reaction of its clones, obtained by somatic embryogenesis. Reproduction of nematodes in clones were compared with an open-pollinated progeny from the same mother plant, C. arabica cultivar Mundo Novo IAC 515-20, susceptible to nematodes, and Clone IAC 1, also susceptible and obtained by somatic embryogenesis. In the last three decades, interest has turned to in vitro cell culture in different areas of coffee research. In vitro techniques have been applied not only for coffee improvement through genetic transformation but also to study various aspects in coffee cells such as chemical (caffeine synthesis and the production of coffee aroma), physiological and more recently, biochemical aspects. The most important advances obtained to date on in vitro coffee techniques in fields like biochemistry, physiology, regeneration systems and genetic engineering, are presented and discussed.

Effect of carbon source on morphology and histodifferentiation of Araucaria angustifolia embryogenic cultures BABT

The aim of the present work was to establish in vitro conditions for the induction, stabilization and proliferation of embryogenic cultures of A. angustifolia. Pre-cotyledonary staged zygotic embryos inoculated BM medium supplemented with 5 µM 2,4- D, 2 µM BAP and Kin, and 3% maltose or sucrose resulted in 66.7% induction rate. The rate of induction of embryogenic cultures was affected by the carbon source, as well the multiplication and morphology of the embryogenic cultures. The first step in plant transformation using somatic embryogenesis or organogenesis for plant regeneration in vitro has been the identification of genotypes with the best totipotency. Thus, the objective of this experiment was to evaluate the totipotency of ten cultivars and one soybean line from the Coodetec Gene Bank. The somatic embryos developed from immature cotyledons measuring from 4 to 6 mm in length, and the organogenesis was evaluated from mature soybean seeds.
